Abstract: The activation of mTORC1 is the driving force of protein synthesis in muscle. However, how to induce intracellular lysine concentration changes, the mechanism of initiation of protein synthesis is unclear. In this project, the porcine skeletal muscle satellite cells is used as a model to expose and screen the key target of mTORC1-sensitive lysine concentration by supposing lysine deficiency and using iTRAQ proteomics technology. The overexpression or interference model is constructed by using CPISPR/Cas9 to analyze the target protein function. Combined with the experience card, the mechanism of mTORC1 induction lysine and activation of protein synthesis is clarified to provide the basis for the regulation of muscle growth.
